Abstract
In this study, the cytotoxic activity of the benzimidazole-based N-heterocyclic carbene (NHC) ligand and the silver(I) (Ag(I)) complex of the ligand were determined by 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) assay on the PC-3 human prostate cancer cell line. In addition, the ligand and complex were tested for selectivity between cancer and healthy cells by applying to human healthy lung fibroblast cells, WI-38. The cells were seeded at a density of 1x105 cells/ml in 96-well plates and incubated with different concentrations (1-20 µM) of ligand and complex for 24, 48 and 72 hours. MTT analysis results showed that the ligand (la) has lower cytotoxic activity on cancer cells than Ag(I)-NHC complex (2a). Also, the benzimidazole ligand (1a) and Ag(I)-NHC complex (2a) showed lower cytotoxic activity against WI-38 human healthy lung fibroblast cells than PC-3 human prostate cancer cells.
